Lipid monolayers: action
نویسنده
چکیده
The activity of phospholipase A on phosphatidyl choline and phosphatidal choline spread as monolayers on phosphate buffers containing snake venom (Crotalus atrox or Naja naja) was studied by measuring the fall of surface potential as a function of time, pH, film pressure, temperature, and concentrations of phosphate and venom. At 25OC, pH 7.0, and 0.2 fig of venom per ml, optimal activity was observed with both venoms on both substrates at 12 dynes/cm film pressure on 0.04 M phosphate. Under these conditions, the pH optimum for C. atrox was broad (6.6-7.4) and that for N . naja was sharp (8.0) for the action on phosphatidyl choline, whereas both venoms had a sharp optimum at pH 8.0 in their action on phosphatidal choline. The optimal temperature with phosphatidyl choline was 27.5OC for N . naja and 4OoC for C. atrox. In line with studies of phospholipase A activity in bulk phase in ether, phosphatidal choline was attacked much more slowly than phosphatidyl choline by C. atrox. Under conditions where both venoms had equal activity on phosphatidyl choline, C. atrox was only half as active as N . naja on phosphatidal choline. The studies suggest that the linkage of the hydrophobic chains in glycerophosphatides may affect their interaction with proteins.

The Action of Some Hemolysis Accelerators upon Lipid and Protein Monolayers
The effect of three accelerators of hemolysis-acetylphenylhydrazine, 9-aminoacridine, and phenothiazone-upon protein and lipid monolayers has been examined. These compounds, in low concentration, cause marked expansion of monolayers of plasma albumin, but have no apparent effect upon cholesterol films. It is suggested that these accelerators may affect the protein component of the erythrocyte m...
متن کاملEffect of tetracaine on DMPC and DMPC+cholesterol biomembrane models: liposomes and monolayers.
Different types of lipid bilayers/monolayers have been used to simulate the cellular membranes in the investigation of the interactions between drugs and cells. However, to our knowledge, very few studies focused on the influence of the chosen membrane model upon the obtained results. The main objective of this work is to understand how do the nature and immobilization state of the biomembrane ...
متن کاملEffects of sodium salts of lyotropic anions on low-temperature, ordered lipid monolayers.
Electrolytes are known to impart considerable disorder to lipid assemblies, including monolayers at the air-water interface, bilayers, and vesicles. In the present work, we have investigated the disordering effect of sodium salts of monovalent anions that span the lyotropic series on the monolayers of 1,2-dipalmitoyl-sn-glycero-phosphocholine (DPPC) at 12 °C. Pressure-area isotherms, Brewster-a...
متن کاملLocal anesthetic inhibition of pancreatic phospholipase A2 action on lecithin monolayers.
Using quantitative data previously reported for the penetration of local anesthetics into lecithin monolayers, the effects of surface and subphase concentrations of anesthetics on the inhibition of pancreatic phospholipase A2 action on didecanoyl phosphatidylcholine monolayers was investigated. Inhibition as a function of subphase concentration of anesthetic was in the order: dibucaine greater ...
متن کاملLateral pressure profiles in lipid monolayers.
We have used molecular dynamics simulations with coarse-grained and atomistic models to study the lateral pressure profiles in lipid monolayers. We first consider simple oillair and oil/water interfaces, and then proceed to lipid monolayers at air/water and oil/water interfaces. The results are qualitatively similar in both atomistic and coarse-grained models. The lateral pressure profile in a ...
متن کاملInsertion of the amyloid precursor protein into lipid monolayers: effects of cholesterol and apolipoprotein E.
APP (amyloid precursor protein), together with Chol (cholesterol) and ApoE (apolipoprotein E), has been linked to Alzheimer's disease. We have examined the hypothesis that interaction of APP with the lipid membranes is modulated by Chol and ApoE. Insertion of APP into lipid monolayers was first evidenced as an increase in the surface pressure. APP injected into a subphase induced a substantial ...
